Can I ask questions in the audition room?

Absolutely! No question is a dumb question. Ask to give yourself clearer direction, which means you will do a better job. I don't think actors should ask questions for the sake of asking a question. There are some casting directors who are okay with no questions. Other casting directors love it when actors ask questions. Bottom line, make sure you are clear. An idea of questions might be, "What is my relationship to the other character(If the information is not provided)?" "How long has my character and the other character known each other?" "What city does the scene take place?" and so forth...
